Biochem/physiol Actions
AKAP1 (A-kinase anchoring protein 1) plays an essential role in the synthesis of non-coding and coding RNA, by directly binding to the RNA. It plays an essential role in steroidogenesis, by anchoring the STAR (steroidogenic acute regulatory) mRNA to mitochondria. Thus, it influences the synthesis of STAR protein and steroids. During G1 phase, this protein helps maintain the integrity of nucleus and hence, cell survival, by controlling PP1 (protein phosphatase1) activity at the nuclear envelope.

AKR1A1 (Aldo-keto reductase family 1, member A1) is associated with the regulation of liver development and hepatocyte maturation. In response to liver damage or partial hepatectomy, it plays an important role in liver regeneration by acting as a hepatotrophic growth factor. AKR1A1 also participates in metabolism of several chemotherapy drugs. It converts anthracyclines doxorubicin (DOX) and daunorubicin (DAUN) to their alcohol metabolites. It has been suggested that AKR1A1 can be used as a genetic biomarker in DAUN-induced cardiotoxicity.

Specificity

RAC-alpha serine/threonine-protein kinase (UniProt: P31749; also known as EC:2.7.11.1, Protein kinase B, PKB, Protein kinase B alpha, PKB alpha, Proto-oncogene c-Akt, RAC-PK-alpha, Akt) is encoded by the AKT1 (also known as PKB, RAC) gene (Gene ID: 207) in human. Akt, a serine/threonine kinase, is a critical enzyme in several signal transduction pathways involved in cell proliferation, apoptosis, angiogenesis, and diabetes. In mammals three isoforms of Akt have been reported (Akt 1, 2, 3) that exhibit a high degree of homology, but differ slightly in the localization of their regulatory phosphorylation sites. Akt1 plays an important role in growth and anti-apoptosis; Akt2 acts primarily as a regulator of glucose metabolism. Although Akt3 does not contribute significantly to the maintenance of normal metabolism in tissues but is essential for the attainment of normal organ size, probably influencing both cell size and number via mTOR activation. Each Akt isoform is composed of three functionally distinct regions: An N-terminal pleckstrin homology (PH) domain that provides a lipid-binding module to direct Akt to PIP2 and PIP3, a central catalytic domain, and a C-terminal hydrophobic motif. Activation of Akt involves growth factor binding to a receptor tyrosine kinase and activation of PI 3-kinase that phosphorylates membrane bound PIP2 to generate PIP3. Activated leukocyte cell adhesion molecule (ALCAM) mediates cell-cell adhesion, either through heterophilic (ALCAM-CD6) or homophilic (ALCAM-ALCAM) interactions. Hemophilic interactions aid in cell migration and invasiveness of tumor cells. ALCAM present on dendritic cells, induces T-cell activation by interacting with cluster of differentiation 6 (CD6) present on T-cells. This heterophilic interaction is necessary for the stabilization of immunological synapses. It mediates the extravasation of leukocytes and their transport to the central nervous system, as well as axonal fasciculation. Colorectal cancer patients, who show increased levels of ALCAM shedding, have poor prognosis and survival rates. The soluble form of ALCAM is related to the aggressive type II phenotype of epithelial ovarian cancer, and can be used as a marker for the same.

Aldehyde dehydrogenase 18 family member A1 (ALDH18A1) is responsible for the reduction of glutamate to 1-pyrroline-5-carboxylate which is an important step in the de-novo biosynthesis of proline, arginine and ornithine. It is a bifunctional enzyme having both -glutamyl kinase (-GK) and -glutamyl phosphate reductase (-GPR) activities. The enzyme undergoes alternative splicing to give rise to two isoforms. The short isoform is active in the gut and it takes part in arginine biosynthesis. The long isoform is involved in the synthesis of proline from glutamate and is expressed in many tissues.

Retinal dehydrogenase 1 (UniProt: P00352; also known as RALDH 1, RalDH1, ALDH-E1, ALHDII, Aldehyde dehydrogenase family 1 member A1, Aldehyde dehydrogenase, cytosolic) is encoded by the ALDH1A1 (also known as ALDC, ALDH1, PUMB1) gene (Gene ID: 216) in human. Retinal dehydrogenase 1 is a ubiquitously distributed cytosolic enzyme that serves as a detoxification enzyme and generates retinoic acid. It catalyzes the oxidation of retinal to retinoic acid that is essential for differentiation, development, and survival of dopaminergic neurons. It can also play a role in cellular defense against oxidative stress by oxidizing 4-hydroxynonenal (4-HNE) produced during lipid peroxidation. It is also involved in several other biological processes, such as stem cell maintenance, UV-radiation resistance, melanogenesis, and spermatogenesis, by catalyzing the conversion of specific aldehydes to their corresponding carboxylic acids. Retinal dehydrogenase 1 contains multiple NAD binding sites. Biochem/physiol Actions
ALDH4A1 (aldehyde dehydrogenase 4 family member A1) catalyzes the oxidation of -glutamate semialdehyde to l-glutamate, in an NAD+-dependent manner. It also catalyzes the second step of hydroxyproline catabolism in humans. Mutations in this gene are linked to type II hyperprolinemia (HPII), which is an autosomal recessive disorder, and results in increased levels of ALDH4A1 enzyme and proline in cerebrospinal fluid (CSF), urine and plasma. HPII is characterized by neurological aberrations including seizures, and mental retardation.

Biochem/physiol Actions
ALDH7A1 (aldehyde dehydrogenase 7 family member A1) is needed for lysine catabolism. It is responsible for NAD+ (nicotinamide adenine dinucleotide)-dependent oxidation of -aminoadipate semialdehyde (AASA) to -aminoadipate (AA). It is also required for the degradation of acetaldehyde which is made during alcohol metabolism. It is upregulated in various cancers, including prostate cancer, non-small cell lung carcinoma and ovarian tumors. Mutations in ALDH7A1 are linked with seizure disorder pyridoxine-dependent epilepsy (PDE).

Aldolase B (ALDOB) is a major enzyme that catalyzes methylglyoxal (MG) formation from fructose in endothelial cells. It blocks high glucose-activated metabolic and signaling pathways by restricting MG production in endothelial cells. The siRNA knockdown of ALDOB in endothelial cells inhibits high glucose-induced MG overproduction. Mutation in ALDOB gene causes hereditary fructose intolerance (HFI).

Fructose-bisphosphate aldolases catalyze the reversible aldol cleavage of fructose-1,6-(bis)phosphate to dihydroxyacetone phosphate (DHAP) and fructose 1-phosphate to either glyceral-dehyde-3-phosphate (G3P) or glyceraldehyde. Aldolase C has been implicated in cell growth, vertebrate development, and other metabolic processes. Its expression is upregulated by nerve-growth-factor-induced B factor, a transcription factor of the thyroid/steroid/retinoid nuclear receptor gene family, suggesting that this isozyme may also have a neurodevelopmental role.

Target description
Fructose-bisphosphate aldolase C (UniProt: P09972; also known as EC:4.1.2.13, Brain-type aldolase) is encoded by the ALDOC (also known as ALDC) gene (Gene ID: 230) in human. Aldolases are glycolytic enzymes that catalyze the reversible conversion of fructose 1-6-diphosphate to glyceraldehyde 3-phosphate and dihydroxy-acetone phosphate via the glycolytic pathway. Within the cell, aldolase is localized in both the cytoplasm and nucleus. In vertebrates, three forms of this aldolases have been described, aldolase A in muscle, aldolase B in liver, and aldolase C in brain. Aldolase C is a homotetrameric enzyme that is expressed mainly in cerebellum and retina. Its kinetic properties are somewhat intermediate between those of the A and B isoforms. In fetal tissues only low levels of aldolase C have been detected. It serves as a useful marker for the study of cerebellar compartmentalization, due to its specific expression in distinct subpopulations of cerebellar Purkinje cells. Activin receptor type-1C (UniProt: Q8NER5, also known as EC:2.7.11.30, Activin receptor type IC, ACTR-IC, Activin receptor-like kinase 7, ALK-7) is encoded by the ACVR1C (also known as ALK7) gene (Gene ID: 130399) in human. ALK-7 is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-20) that is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 21-113), a transmembrane domain (aa 114-134, and a cytoplasmic domain (aa 135-493). ALK-7 expression is reported in pancreas, heart, colon, small intestine, ovary and the hippocampus, medulla oblongata and putamen of the brain. It serves as a transmembrane type IC receptor serine kinase for activin or activin-like ligands. The receptor complex consists of 2 type II and 2 type I transmembrane serine/threonine kinases. Type II receptors phosphorylate and activate type I receptors which auto-phosphorylate, then bind and activate SMAD transcriptional regulators, SMAD2 and SMAD3. The type I receptors contains a Ser/Thr kinase domain in its cytoplasmic domain (aa 194-485) that is essential for signaling, while the type II are receptors are essential for ligand binding. ALK-7 is reported to negatively regulate glucose-stimulated insulin release in -cells. Mice lacking ALK-7 display normal pancreas organogenesis but develop an age-dependent syndrome involving progressive hyperinsulinemia, reduced insulin sensitivity, liver steatosis, impaired glucose tolerance, and islet enlargement.
