Biochem/physiol Actions
Claudin-1 (CLDN1) plays a key role in the tight junction barrier formation. It interacts with claudin-3 and modulates tight junctions in human airway epithelium. CLDN1 acts as a receptor for the hepatitis C virus entry and participates in the internalization of the virus. Mutation in CLDN1 gene is implicated in ichthyosis and neonatal sclerosing cholangitis (NISCH syndrome). CLDN1 is highly expressed in colon cancer.

CLDN11 (Claudin-11) gene encodes a protein belonging to the claudin family. Claudin-11 is a tight junction protein human blood-testis barrier (BTB). It also forms a component of the myelin in the CNS and functions in the proliferation and migration of oligodendrocytes. Dsyfunction of this gene has been implicated in disruption of BTB. Claudins play an imporatnt role in compartmentalization in multicellular organisms.

Biochem/physiol Actions
CLDN7 (claudin-7) prevents the invasion and migration of human lung cancer cells, and induces apoptosis by activating caspases, post cisplatin treatment of lung cancer cells. It interacts with CD44 and EpCAM, and might form a membrane-anchoring protein complex, which helps it to reside in the plasma membrane. It is up-regulated in colon cancer, where it facilitates tumorigenesis. Its expression is also associated with inflammatory bowel disease (IBD) and colitis-associated cancer. It is either completely inactivated or under-expressed in human breast cancer, and is also linked with relapse of the same. Studies on human breast cancer MCF-7 cell line, show that tumor necrosis factor (TNF)α induces the expression of CLDN7, which leads to accumulation of β-catenin in the cytosol. This way CLDN7 regulates apoptosis of MCF-7 cell line. It is over-expressed in epithelial ovarian cancer, where it might be involved in its invasion.

Poly [ADP-ribose] polymerase 1 (UniProt: P09874; also known as EC:2.4.2.30, PARP-1, ADP-ribosyltransferase diphtheria toxin-like 1, ARTD1, DNA ADP-ribosyltransferase, PARP1, NAD(+) ADP-ribosyltransferase 1, ADPRT 1, Poly[ADP-ribose] synthase 1, Protein poly-ADP-ribosyltransferase PARP1) is encoded by the PARP1 (also known as ADPRT, PPOL) gene (Gene ID: 142) in human. PARP1 is zinc-dependent DNA binding protein that recognizes DNA strand breaks and plays a role in DNA repair. It is involved in the base excision repair (BER) pathway by catalyzing the poly(ADP-ribosyl)ation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. This modification follows DNA damages and appears as an obligatory step in a detection/signaling pathway leading to the reparation of DNA strand breaks. In addition to BER pathway, it is also involved in double-strand breaks (DSBs) and accumulates at DNA damage sites and promotes homologous recombination repair by mediating poly-ADP-ribosylation. PARP1 also mediates the poly(ADP-ribosyl)ation of a number of proteins and mainly mediates glutamate and aspartate ADP-ribosylation of target proteins. The ADP-D-ribosyl group of NAD+ is transferred to the acceptor carboxyl group of glutamate and aspartate residues and further ADP-ribosyl groups are transferred to the 2′-position of the terminal adenosine moiety, building up a polymer with an average chain length of 20-30 units. The DNA-binding region of PARP1 is localized to amino acids 2-373. It also contains two zinc-finger regions (aa 9-93 and 113-203). PARP is cleaved between amino acids Asp214 and Gly215 to yield two fragments: a 29 kDa (N-terminal) and a 85 kDa (C-terminal DNA-binding domain). The carboxyl-terminally located 54-kDa domain of PARP represents the NAD1-binding domain with the characteristic PARP signature, which includes a highly conserved sequence comprising the catalytically crucial amino acid residue Glu-988. Its catalytic activity is stimulated by noncovalent contact of the DNA-binding domain with DNA strand breaks and results in the post-translational modification of various acceptor proteins. In between the DNA binding domain and the NAD1-binding domain, it also contains a 22-kDa auto-modification domain. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. CLPS (colipase) functions as a cofactor for pancreatic triglyceride lipase for the efficient hydrolysis of dietary lipid. Its expression is increased by secretin and dietary fats. Arg92Cys polymorphism in this gene is linked with increased susceptibility to type II diabetes. This polymorphism reduces functionality against long-chain triglycerides, which might be a possible cause of type II diabetes.

Target description
Clusterin (UniProt: P10909; also known as Aging-associated gene 4 protein, Apolipoprotein J, Apo-J, Complement cytolysis inhibitor, CLI, Complement-associated protein SP-40,40, Ku70-binding protein 1, NA1/NA2, Sulfated glycoprotein 2, SGP-2, Testosterone-repressed prostate message 2, TRPM-2) is encoded by the CLU (also known as APOJ, CLI, KUB1, AAG4) gene (Gene ID: 1191) in human. Clusterin is a ubiquitously expressed protein that functions as extracellular chaperone that prevents aggregation of non-native proteins. It also prevents stress-induced aggregation of plasma proteins. Clusterin is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to generate the mature form that can be proteolytically cleaved into a b-chain (aa 23-227) and an a-chain (aa 228-449). However, proteolytic cleavage is not necessary for its chaperone activity. Six different isoforms of Clusterin have been described that are produced by alternative splicing. Isoform 1 is shown to retro-translocate from the secretory compartments to the cytosol upon cellular stress, but isoforms 4 and 6 retain their cytoplasmic localization in stressed and unstressed cell. It maintains partially unfolded proteins in a state appropriate for subsequent refolding by other chaperones, such as HSPA8/HSC70, but does not refold proteins by itself. Clusterin is reported to bind to Amyloid b-peptides and alters their aggregation and promote their clearance, suggesting its neuroprotective role. Physical form
Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
