XYLT1 (xylosyltransferase I) catalyzes the O-glycosylation of serine residues of proteins by the transfer of UDP-xylose. This step is important in the initiation of the glycosaminoglycan (GAG) chains during proteoglycan synthesis. Defects in this gene can cause an autosomal recessive short stature syndrome and Desbuquois Dysplasia Type 2.

Yes-associated protein 1 (YAP1) interacts with the src family kinase c-Yes by its SH3 binding motif and with protein 73 using WW motif. YAP promotes cell and tissue growth. YAP1 interacts with Receptor tyrosine-protein kinase (ErbB4) receptor and regulates transcription. YAP is oncogenic and contributes to tumor progression. Phosphorylation of YAP is critical for regulating apoptosis. YAP1 levels are elevated in human cancers. It is highly expressed in hedgehog-associated medulloblastomas. In bacteria, YBEY (putative ribonuclease) is involved in protein synthesis. Escherichia coli cells with YBEY deletion exhibit defects in ribosome assemble and activity, as well as in attenuation of 16S, 23S and 5S rRNA processing. Biochem/physiol Actions
YIF1A (Yip1 interacting factor homolog A) is a Yip1p interacting protein, with which it forms a heteromeric complex. This complex is essential for the binding of endoplasmic reticulum (ER)-derived vesicles with Bos1p and Sec22p proteins of SNARE complex. It might be involved in maintaining the structure of Golgi apparatus. It is also an interacting partner of vesicle-associated membrane protein (VAMP) associated protein B (VAPB), which is an essential part of the early secretory pathway. Both these proteins play essential roles in ER-Golgi transport, and errors in YIF1A sorting might be a contributing factor to VAPB-associated motor neuron disease. It is also essential for the normal growth of dendrites. It has a ubiquitous expression in the central nervous system (CNS). Studies in mice show that this protein could be one of the factors implicated in amyotrophic lateral sclerosis (ALS).

Biochem/physiol Actions
ATP-dependent zinc metalloprotease YME1L1 protects the mitochondria, regulates the movement of proteins across the mitochondrial membrane and the proteolysis of misfolded proteins. It also controls the mitochondrial dynamics. YME1L1 controls the formation of respiratory chain molecules by proteolysis and maintains the cristae structure. YME1L1 helps mitochondria to adjust to different types of stress. In humans, mutations in YME1L1 leads to mitochondriopathy with optic nerve atrophy and mitochondrial network fragmentation. Biochem/physiol Actions
YWHAQ (tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation protein, θ) gene encodes a protein belonging to the 14-3-3 family of proteins that are involved in signal transduction by binding to phosphorylated Ser residues via the two consensus sequences of RXXXpSXP and R(S/X)XpSXP. The functional unit is a dimer and may function in several processes including signal transduction, apoptosis, cell cycle progression and DNA replication. The encoded protein is also called as 14-3-3 θ. It may serve as a potential marker of lung cancer. This protein has been found to be upregulated in amyotrophic lateral sclerosis spinal cord. The promoter of YWHAQ gene has been found to be hypermethylated in preeclamptic placentas.

Tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation protein-ε (YWHAE) binds to cyclin-dependent kinase 5 (CDK5)-phosphorylated nudE neurodevelopment protein 1-like 1 (NUDEL) and maintains its phosphorylation state. NUDEL is a cytoplasmic regulator of neuronal migration. YWHAE is required for neuronal development as it aids in the transport of the NUDEL complex. It has protective and preventive effect on schizophrenia.
