Bob is a novel pre-clinical model for functional studies in CRPC and especially for studying the CRPC "basal" phenotype. SerBob grows as a monolayer culture with limited differentiation. The Y chromosome could not be detected in this cell line by short tandem repeat (STR)-PCR analysis when tested at ECACC. The cell line has a variety of unusual characteristics including the morphology of an undifferentiated large cell variant of small cell lung carcinoma (SCLC) but biochemical properties of a classical SCLC. SHP-77 express neural cell adhesion molecule (NCAM), NHK-1, CD56 and the cell surface leucocyte antigen Leu-7(HNK-1;CD57). Other neuroendocrine markers are L-dopa decarboxylase and dense core secretory granules. Gland formation and intracytoplasmic lamellar bodies have been shown by electron microscopy. This cell line is useful as in vitro target in 51Cr and III In release cytotoxicity assays. They are suitable for in vitro nude mice assays for evaluation of immune reactivity of cells and serum from lung cancer patients. Cell Line Description
SK-GT-2 was established from a primary adenocarcinoma of gastric fundus from a 72-year-old Hispanic male previously treated for squamous cell carcinoma with radiation therapy and radical neck dissection. Pathology of recurrence (1 year) demonstrated ulcerating poorly differentiated adenocarcinoma of the stomach invading spleen and epigastic fat (T4) with metastases to perisplenic and perigastric nodes (N1). SK-GT-2 was found to be tumorigenic in athymic nu/nu mice. The Y chromosome could not be detected in this cell line by short tandem repeat (STR)-PCR analysis when tested at ECACC. It is a known phenomenon that due to the increased genetic instability of cancer cell lines the Y chromosome can be rearranged or lost resulting in lack of detection. The cell line is identical to the source provided by the depositor based on the STR-PCR analysis.

Full recovery may take up to 2 weeks. A centrifugation step to remove the cryoprotectant is essential. Rapidly thaw the frozen ampoule in a water bath at 37°C for 1-2 minutes. Transfer the contents to a centrifuge tube and slowly add 5-10ml of pre-warmed growth media. Remove a sample for counting. Centrifuge at 100g for 2-3 minutes to pellet cells and seed at a relatively high density of 5-7 x 105 cells/ml. Leave culture flask upright and observe regularly until viable proliferating cells are seen.
