It is used for microbiological validation of aseptic filling lines across the beverage industry because it creates optimum growth conditions for fungi and bacteria at various pH levels. The pH of the medium can be adjusted to the filling line criteria. At a pH of 4.3, fungi and bacteria which are adapted to grow under high acid conditions show visible growth within 7 days of incubation at 28°C. Bacteria that grow best at neutral to slightly alkaline pH, like Bacillus subtilis, show no growth/very limited growth at pH 4.3. Any growth within the medium (turbidity), on the surface of the medium, or the inner wall or bottom of the tubes and bottles indicates the presence of spoilage organisms. Experiments have shown that the medium could also be used after freezing and thawing, thus making this medium suitable for ice crusher installations.ApplicationLinden Grain Medium is suitable for the detection of yeasts, molds, and bacteria in beverages. eCl@ss: 32160801 NACRES: NA.74 General descriptionm-Green Yeast and Mold Broth is a relatively more complex formula compared to other media used for isolation of fungi and yeast.{10} This formulation is rich in nutrient which allows excellent fungal growth. Bacterial growth is inhibited by an acidic pH. Enzymatic digest of casein and enzymatic digest of animal tissue provide nitrogen, carbon, and amino acids in m-Green yeast and mold Broth. Yeast extract is the vitamin source. Dextrose is an energy source for metabolism of fungi. Potassium phosphate is a buffering agent. Magnesium sulphate, Thiamine, and Diastase (a mixture containing amylolytic (starch) enzymes) provide essential ions, minerals, and nutrients. Bromocresol Green is the pH indicator, facilitating visualization and counting of fungal colonies. The colonies are green due to the diffusion of bromocresol green into the colonies (alkaline reaction). Acidic end products from the colonies diffuse into the medium, further reducing the pH and causing the dye to turn yellow (acid reaction). Yeast produces green, opaque large colonies.
