Most of the Escherichia coli strains can be differentiated from other coliforms by the presence of enzyme glucuronidase which is highly specific for Escherichia coli. Escherichia coli cells absorb X-glucuronide and the intracellular glucuronidase splits the bond between the chromophore and the glucuronide. The released chromophore gives the blue coloration of the colonies.Preparation NoteSuspend 36.6 g per litre in distilled water. Sterilize by autoclaving at 121°C for 15 minutes. The ingredients help even the sublethally injured coliforms to grow rapidly, Tergitol™ inhibits Gram-positive as well as some Gram-negative bacteria other than coliforms. The chromogenic mixture contains two chromogenic substrates as Salmon-GAL and X-glucuronide. The enzyme -D-galactosidase produced by coliforms cleaves Salmon-GAL, resulting in the salmon-to-red coloration of coliform colonies. The enzyme -D-glucuronidase produced by E. coli, cleaves X-glucuronide. E. coli forms dark blue-to-violet colored colonies due to cleavage of both Salmon-GAL and X-glucuronide. The addition of tryptophan improves the indole reaction. To confirm E. coli, add a drop of Kovacs reagent (Catalog No. 60983) on the dark blue-to-violet colony. Formation of cherry-red color indicates the positive reaction.Preparation NoteSuspend 26.5 g in 1 litre distilled water. Heat in a boiling water bath or in a free flowing steam, with stirring to dissolve the medium completely (approximately 35 minutes). Medium may show haziness, but it does not affect the performance. If desired to inhibit Pseudomonas and Aeromonas species add 5 mg Cefsulodin (Cat. No. 22126) for surface and pour plate method and 10 mg Cefsulodin for membrane filter technique. The detection of the -D-glucosidase activity is possible by the addition of aesculin and ammonium iron (III) citrate, forming a black complex of aesculin with iron (III) ions. A black coloration can develop during incubation, but this reaction is not exclusive to Listeria spp. and cultures that show no blackening cannot be assumed to be free of Listeria spp. According to EN ISO 11290-1, every primary and secondary enrichment in Fraser broth has to be sub-cultured on selective plating media. Generally all ferrioxamines are trihydroxamates and the deferrioxamines are either linear or cyclic (D2 and E).

Ferrioxamine E from Streptomyces antibioticum is a siderophore which facilitates the supply of iron (III), an essential trace element, to bacteria involved in food poisoning, including Salmonella, Enterobacter saka-zakii and Yersinia enterocolitica. It promotes rapid growth by reducing the lag phase in culture media and reactivates dormant bacteria. Use of ferrioxamine E can permit rapid detection of low bacterial cell counts, even from dry products like spices and tea after long storage periods. It is an essential component of quality control efforts in the food industry.
Ferrioxamine E is often used in Buffered Peptone Water, the medium recommended by the ISO-Norms for Enterobacteriacea. Ferrioxamine E also improves the motility of Salmonella, which helps to improve the identification by semisolid selective motility media like SIM, MRSV, DIASSALM or SMS. Ferrioxamine E does not improve growth of Escherichia coli, Shigella, Proteus, Providencia and Morganella species; a feature that makes it a semi-selective compound.
