Biochem/physiol Actions
Tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) activates the extrinsic apoptotic pathway by binding death receptors (DR)-4 and 5. In addition to DR 4 and 5, TRAIL activates several other receptors such as decoy receptor (DcR)1, DcR2 and a soluble receptor called osteoprotegerin. TRAIL plays a vital role in the inhibition of autoimmune inflammation and cell cycle progression. TRAIL selectively kills tumor cells without damaging normal tissues and therefore, it might act as a tumor-selective anticancer agent. TRAIL-induced apoptosis is involved in various cellular processes including activation-induced cell death, homeostasis, intrathymic negative selection, autoimmunity suppression, and immune surveillance. Biochem/physiol Actions
Tumor necrosis factor (TNF) superfamily member 4 (Tnfsf4) signals specifically through the OX40 receptor, which is expressed primarily on CD4+T cells but also on certain activated CD8+T cells. Tnfsf4 acts as a costimulatory signal, which is essential for a productive interaction between antigen presenting cells and their target T-cells. It enhances cell proliferation and survival and increases expression of regulated on activation, normal T cell expressed and secreted (RANTES) or chemokine ligand 5 (CCL5), interleukins-2 and -3 and interferon (IFNg). Tnfsf4 signaling plays an important role in immuno-regulatory communication and immune-tolerance. Biochem/physiol Actions
Tyrosine-protein kinase receptor (TYRO3) is a transmembrane tyrosine kinase receptor. It plays a significant role in the brain development during embryogenesis. It plays an important role in apoptotic cell clearance. In hepatocellular carcinoma, it is associated with tumor growth, aggressiveness and liver dysfunction. Mutations in the TYRO3 gene intronic regions are linked with asthma. TYRO3 is also a suppressor of type 2 immunity. As part of the TAM (TYRO3, AXL and MER receptors) family, it is activated by PROS1 (protein S) and GAS6 (growth arrest-specific gene 6). Changes in the expression of TYRO3 are linked with the proliferation and survival of cancer cells. Biochem/physiol Actions
The members of epidermal growth factor receptor (EGF R) or the ErbB receptor family have been identified as useful biomarkers and targets for cancer therapy. EGF R binds EGF and induces tyrosine phosphorylation leading to proliferation of cells. EGF R is capable of binding transforming growth factor-α and heparin-binding EGF in addition to EGF. There are numerous effector molecules activated by EGF R that result in a variety of biological processes such as morphogenesis, cell motility, apoptosis, differentiation and organ repair and maintenance. Deregulation of EGF R signaling is implicated in progression of a wide variety of tumors, invasion and metastasis. Biochem/physiol Actions
EGFR (epidermal growth factor receptor) functions as a receptor for EGF-family of proteins, which are peptide growth factors. This receptor-ligand system is also involved in the pathogenesis and progression of various types of carcinomas. In cancer, EGFR pathways promotes cellular proliferation, angiogenesis, metastasis and apoptosis. The microRNA miR-218-5p interacts with and controls EGFR, and in non-small cell lung cancer (NSCLC), the levels of EGFR are inversely related to miR-218-5p. miR-218-5p inhibits proliferation and migration of lung cancer cells by binding with EGFR, and thus, has potential in future NSCLC therapies.
