CXCR-4 is a receptor for the C-X-C chemokine CXCL12/SDF-1 that transduces a signal by increasing intracellular calcium levels and enhancing MAPK1/MAPK3 activation. CXCR-4 is expressed in numerous tissues and two isoforms have been reported that are generated by alternative splicing. Isoform 1 is the predominant form in all tissues tested. CXCR-4 is rapidly phosphorylated on serine and threonine residues in the C-terminal upon agonist stimulation. Phosphorylation at Ser324 and Ser325 leads to recruitment of ITCH (E3 ubiquitin-protein ligase), ubiquitination and protein degradation. Sulfation on Tyr21 is shown to be essential for efficient binding of CXCL12/SDF-1alpha and it promotes its dimerization. Tyr7 and Tyr12 are sulfated in a sequential manner following complete sulfation of Tyr21. CXCR-4 acts as a co-receptor for human HIV-1 virus isolates (CD4 being the primary receptor) and as a primary receptor for HIV-2 isolates. CXCR-4 can also bind bacterial lipopolysaccharide (LPS) and mediate LPS-induced inflammatory response, including TNF-alpha secretion by monocytes. Mutations in CXCR4 gene cause WHIM syndrome that is characterized by neutropenia, hypogammaglobulinemia and extensive human papillomavirus (HPV) infection.

These proteins are the regulatory subunits of the cell cycle-dependent kinases that are responsible for the phosphorylation of several cellular targets. The B cyclins, when combined with p34cdc2, a catalytic subunit of the cell cycle dependent kinase, form an enzyme that is active at the transition to the G2 phase, attaining maximum level at M phase. Anti-Cyclin B1 may be used for the detection of cyclin B1 expression and for measuring relative differences in cyclin B1 level as a function of cell cycle phase.

The MT-CYB gene is a good phylogenetic marker. Defects in MT-CYB gene are a rare cause of mitochondrial dysfunction underlying different myopathies, which include mitochondrial encephalomyopathy, hypertrophic cardiomyopathy, and sporadic mitochondrial myopathy. Cytokeratin-18 is a type I, acidic, protein that is expressed in colon, placenta, and liver. Higher expression levels have been reported in lymph nodes of breast carcinoma. Cytokeratin-18 is involved in the uptake of thrombin-antithrombin complexes by hepatic cells. Upon phosphorylation, it plays a role in filament reorganization. It is proteolytically cleaved by caspases during epithelial cell apoptosis and this cleavage is shown to occur at Asp-238. Dab 1 is mainly expressed in the brain and serves as an adapter molecule involved in neural development where it is reported to direct the migration of cortical neurons past previously formed neurons to reach their proper layer. It is a cytosolic protein that is shown to be essential component of the reelin signaling pathway. It is shown to bind to the cytoplasmic tail of Reelin receptor, APO E receptor 2, and to LDL receptor. Dab1 can be phosphorylated on Tyr198, Tyr220, and Tyr232 upon reelin induction in embryonic neurons. It can also be phosphorylated on Ser524, but this phosphorylation is independent of reelin signaling. Tyrosine phosphorylation of Dab1 by Src family tyrosine kinases is considered to be the most critical downstream event in Reelin signaling. Dab1 contains a phosphotyrosine interaction domain (PID) (aa 36-189), which specifically binds to the Asn-Pro-Xaa-Tyr(P) motif found in many tyrosine-phosphorylated proteins. Dach1 is a transcription factor that is involved in regulation of organogenesis. Four isoforms of Dach1 have been reported that are produced by alternative splicing. It is shown to be widely expressed. Isoform 2 is found in brain, heart, kidney, liver, leukocytes and spleen. Isoform 3 is found in liver and heart and isoform 4 is found in spleen. Clone 1F6.1 exhibits reactivity with all four isoforms. Dach1 is reported to act as regulator of SIX1, SIX6, and probably SIX5. It also acts as a a corepressor of SIX6 in regulating proliferation by directly repressing cyclin-dependent kinase inhibitors, including the p27Kip1 promoter. Dach1 is shown to Inhibit TGF-beta signaling through interaction with SMAD4 and NCOR1. It binds to chromatin DNA via its DACHbox-N domain, which forms a structure containing a DNA binding motif similar to that of the forkhead/winged helix domain. PELP1 (proline, glutamic acid, and leucine rich protein 1) serves as a Dach1-binding protein and the carboxyl terminus of Dach1 is required for binding to PELP1. Dach1 is reported to prevent the proliferation of cancerous cells due to being a natural repressor of estrogen receptors (ER) in the normal function of mammary tissue. DCLK1 expression is reported to be critical for growth of human colon cancer cells and its expression is up-regulated in response to growth factors resulting in hyperproliferation. Both long (L) (~80-82 kDa) and short (S) (~45-50 kDa) isoforms of DCLK1 have been described. The long isoform of DCLK1 contains two N-terminal doublecortin domains, which bind microtubules, a C-terminal serine/threonine kinase domain with homology to CaM kinases, and a middle serine/proline rich domain that mediates protein interactions. The short isoform, DCLK1-S, has been reported in human colorectal cancer cells and in cancer stem cells, while normal colon cells express only the long isoform of DCLK1. The short isoforms lacks the two N-terminal doublecortin domains. Although DCLK1-S differs from the long isoform by only six amino-acids., this mono-specific antibody does not cross-react with the long isoform and is shown to specifically detect cancer stem cells. The short isoform is not a splice variant, but is expressed from an alternate beta promoter in intron V of the gene that results in the presence of the unique sequence of amino acids. These amino acids are absent in the long isoform, which is expressed by the alpha promoter in the 5’ region of the gene. It has been reported that the 5′ (alpha)-promoter of the human DCLK1 gene becomes epigenetically silenced during colon carcinogenesis, resulting in loss of expression of the canonical long isoform in human colorectal adenocarcinomas (hCRCs). Instead, hCRCs express the short isoform, DCLK1-S. Dcp2 contains a functional Nudix (nucleotide diphosphate linked to an X moiety) motif (aa 95-226) that is essential for its decapping activity. Two isoforms of Dcp2 have been described that are produced by alternative splicing. Dcp2 is reported to play a role in replication-dependent histone mRNA degradation. It has higher activity towards mRNAs that lack a poly(A) tail and does not display any activity towards a cap structure lacking an RNA moiety. Dcp2 is predominantly cytoplasmic, however, a small amount may also be found in the nucleus. DDX6 is considered to be a P-bodies marker due to its mediating role between translational repression and P-bodies formation and toits interaction with Ago2 and the RISC complex that also mediates translational silencing by miRNAs.1-4 Viruses like Hepatitis C virus (HCV) or West Nile virus (WNV) utilize the host cell DDX6 to promote virus infection by recruiting it to the viral replication sites where it is involved in stabilization of the viral genome and the shift between translation and replication.2,5-6DDX6 was reported to be overexpressed in colorectal adenocarcinomas, gastric cancer and in most malignant cell lines.7-9 It was suggested that DDX6 acted as an upstream cancerous enhancer through RNA-binding of HER2 and FGFR2 mRNAs that positively regulates post-transcriptional processes in cancer cells.7

Del-1 is a calcium-dependent glycoprotein that is secreted by endothelial cells and is shown to be expressed in a developmentally restricted pattern during embryogenesis. In adult tissues it is expressed in endothelial cells and in macrophages. Del-1 is reported to prevent apoptosis in endothelial cell subjected to TNF alpha, etoposide, and anoikis treatment. This anti-apoptotic effect is shown to be mediated specifically through binding of integrin alphav beta3 by the RGD motif. Del-1 expression is reported to be inversely related to that of IL-17 that is shown to inhibit Del-1 via a GSK-3beta and C/EBPb-dependent mechanism. This inhibitory action can be reversed at the PI3K/Akt signaling induced by D-resolvins. It serves as an endothelial cell-secreted homeostatic factor that regulates local inflammation in several tissues. In adult striated muscle desmin forms a fibrous network connecting myofibrils to each other and to the plasma membrane from the periphery of the Z-line structures. It may also act as a sarcomeric microtubule-anchoring protein and specifically associate with detyrosinated tubulin-alpha chains, leading to buckled microtubules and mechanical resistance to contraction. Desmin is particularly abundant at the myotendinous junctions and at the neuromuscular junctions of skeletal muscle. Mammalian cardiac muscle cells contain higher amounts of desmin (up to 2% of total protein) compared to skeletal muscle cells (0.35% of total protein). Mutations in DES gene have been linked to familial cardiac and skeletal myopathy characterized by skeletal muscle weakness associated with cardiac conduction blocks, arrhythmias, and restrictive heart failure. It can also lead to myofibrillar destruction with intracytoplasmic accumulation of desmin-reactive deposits in cardiac and skeletal muscle cells. (rEF.: Denise Paulin, D., and Li, Z (2004). Desmoplakin is a major high molecular weight homodimeric protein that is involved in the organization of the desmosomal cadherin-plakoglobin complexes into discrete plasma membrane domains and in the anchoring of intermediate filaments to the desmosomes, which are prominent adhesive junctions present between many epithelial cells and cardiomyocytes. Desmoplakin is the most abundant of the desmosomal proteins and plays a key role in tissue morphogenesis and maintaining the integrity of tissue. Desmoplakin is composed of an N-terminal plakin domain, a central alpha-helical coiled-coil rod domain, and a C-terminal intermediate filament-binding domain. The N-terminal domain is required for desmosome association and the C-terminal binds to desmin. Three isoforms for Desmoplakin have been reported that are produced by alternative splicing. Desmoplakin 2 and 3 lack most of the central rod domain. Desmoplakin 1 is the dominant isoform in the cardiac tissue. Desmoplakin is phosphorylated by protein kinase A on serine 2849 and this phosphorylation affects its association with epidermal, simple cytokeratins, and VIM intermediate filaments. Biochem/physiol Actions
DNA fragmentation factor subunit alpha (DFFA) is a component of DNA fragmentation factor (DFF). It is mainly associated with apoptosis and genomic stability. Caspase-3-mediated cleavage of DFFA releases DFF40 (DNA fragmentation factor 40) which degrades chromosomal DNA. During menstruation, the apoptosis of endometrium cells is associated with DFFA. Its expression decreases after menopause. Silencing of DFFA increases doxorubicin effects in breast cancer cells.

Biochem/physiol Actions
DGCR8 (DGCR8 microprocessor complex subunit) participates in the biogenesis of microRNA (miRNA, miR) as a major component of the microprocessor complex. DGCR8 provides guidance to the RNase III enzyme, Drosha during rRNA processing. It forms the microprocessor complex by binding to the RNase III enzyme Drosha, which further converts long primary miRNAs (pri-miRNAs) into short hairpins called precursor miRNAs (pre-miRNAs). The processed hairpins finally are transported to the cytoplasm for further processing by Dicer into mature miRNAs. DGCR8 is also required for global gene regulation and silencing of embryonic stem cell self-renewal. Deletion of DGCR8 chromosomal location has been reported in the DiGeorge and velocardiofacial syndrome.

Biochem/physiol Actions
DGCR8, also known as DiGeorge syndrome critical region 8, DGCRK8 is the cofactor that interacts with drosha and forms a functional complex called the ‘‘Microprocessor” which is essential for microRNA (miRNA) maturation. DGCR8 contains an N-terminal region which is critical for nuclear localization and the C-terminal region stably interacts with the pri-miRNAs.
DGCR8 (DGCR8 microprocessor complex subunit) participates in the biogenesis of microRNA (miRNA, miR) as a major component of the microprocessor complex. DGCR8 provides guidance to the RNase III enzyme, Drosha during rRNA processing. It forms the microprocessor complex by binding to the RNase III enzyme Drosha, which further converts long primary miRNAs (pri-miRNAs) into short hairpins called precursor miRNAs (pre-miRNAs). The processed hairpins finally are transported to the cytoplasm for further processing by Dicer into mature miRNAs. DGCR8 is also required for global gene regulation and silencing of embryonic stem cell self-renewal. Deletion of DGCR8 chromosomal location has been reported in the DiGeorge and velocardiofacial syndrome.

Biochem/physiol Actions
Deleted in liver cancer 1 (DLC1) suppresses cell growth and invasion in cancers and thus functions as a tumor-suppressor gene. It also induces cell apoptosis by controlling cyclin D1 expression. Single-stranded guanine (G)-rich DNA can form stable secondary structures called G-quadruplexes (G-tetrads or G4), which can influence methylation at CpG islands (CGIs). G4 structure is reported to be tightly associated with CGI hypomethylation in the human genome. Four guanine bases can associate through Hoogsteen hydrogen bonding to form a square planar structure called a guanine tetrad (G-tetrad), and two or more G-tetrads can stack on top of each other to form a G-quadruplex. The quadruplex structure is further stabilized by the presence of a cation, especially potassium, which sits in a central channel between each pair of tetrads. A DNA G-tetrad can be formed within one DNA strand (intramolecular), between two DNA strands (bimolecular), or four DNA strands (tetramolecular). G4 DNA can arise anywhere in the genome where sufficiently long stretches of single-stranded G-rich DNA are exposed during replication, transcription or recombination. Chemical analysis of quadruplex-forming oligonucleotides has revealed the existence of a plethora of dynamic quadruplex structures with varying stabilities. DNA helicase B is highly expressed in testis and thymus and weaker expression is observed in liver, spleen, kidney, and brain. It is predominantly nuclear, however, following phosphorylation at serine 967 by CDK2 during the G1/S transition it is exported into the cytoplasm. Following DNA damage, it is recruited to sites of double-strand breaks by the RPA complex. Its recruitment to single-stranded DNA following DNA damage leads to inhibition of nucleases catalyzing resection, such as EXO1, BLM and DNA2, possibly via the 5-3 ssDNA translocase activity of HELB. As cells approach S phase, DNA end resection is promoted by the nuclear export of HELB following its phosphorylation. Its nuclear export signal is localized to amino acids 1055-1078. It uses either magnesium or manganese ions to support its helicase activity and the DNA helicase B activity is inhibited by salt concentrations higher than 100 mM. It binds strongly to single-stranded DNA in the absence of ATP, but dissociates readily in the presence of 1 mM ATP. Two isoforms of DNA helicase B have been reported that are produced by alternative splicing.

DNMT3L is a catalytically inactive regulatory factor of DNA methyltransferases that can either promote or inhibit DNA methylation depending on the context.
It is shown to be essential for the function of DNMT3A and DNMT3B. It activates DNMT3A and DNMT3B by binding to their catalytic domain. It acts by accelerating the binding of DNA and S-adenosyl-L-methionine (AdoMet) to the methyltransferases and dissociates from the complex after DNA binding to the methyltransferases. DNMT3L recognizes unmethylated histone H3 lysine 4 (H3K4me0) and induces de novo DNA methylation by recruitment or activation of DNMT3. In germ cells, it is required for the methylation of imprinted loci together with DNMT3A. In male germ it is essential for the methylation of retrotransposons, which prevents their mobilization. DNMT3A also plays a key role in embryonic stem cells (ESCs) by acting both as a positive and negative regulator of DNA methylation. It promotes DNA methylation of housekeeping genes together with DNMT3A and DNMT3B. It can also serve as an inhibitor of DNA methylation at the promoter of bivalent genes. Biochem/physiol Actions
Zizimin1/ dedicator of cytokinesis 9 (DOCK9) is found to activate Cdc42 through binding to its CZH2 domain. Interestingly, the CZH1 domain of Zizimin1 at the N-terminal binds to its CZH2 domain and thus inhibits Cdc42 binding. The Pleckstrin homology (PH) domain at the N-terminal, binds phosphoinositides and results in zizimin1 membrane targeting.

Storage and Stability
